High-Throughput Oligonucleotide Mapping Workflow Enabled by High-Resolution Ion Mobility

Demonstrate the ability of high-resolution ion mobility (HRIM) to enhance oligonucleotide mapping through high-throughput full scan separations and superior isomer resolution.
Monitoring oligonucleotide sequence coverage and modification presence (including isomeric forms) is critical to ensure safety, efficacy, and quality of synthetic RNA therapeutics. A simple 1-minute FIA-HRIM-MS MAF workflow using automated software for data analysis was demonstrated for a modified sgRNA molecule where 98% sequence coverage was achieved using two enzymes for digestion, in addition to the ability to monitor isomeric PS modifications.
